Abstract: The objectives of this research were 1) to investigate the phonemes of consonants and vowels of Myanmar language used in Yangon City of Myanmar and Samut Sakhon of Thailand, and 2) to compare the similarities and differences of phonemes of Myanmar language used in Yangon City of Myanmar and Samut Sakhon province of Thailand. The study was a qualitative method. The research tools included questionnaire and interviews, and the phoneme testing of 400 Burmese words of pronunciation was implemented as the data presented in the difference and similarity, and the test was verified by 3 experts and used as data analysis. The research results showed that native Burmese speakers acquire another language through communication with friends at fluent and confident level in a particular area in Thailand, and the testing of phonemes was implemented to verify the similarities and differences of 20 categories of Burmese words used in two areas; Yangon City of Myanmar and Samut Sakhon province of Thailand, the phonemes of stops, affricate and affricative consonant sounds, such as /သ/, /θ/, /စ/, /ရွ/, /ʃ/, /ဟ/, /ဓ/, /ð/, /ဆ/, /sʰ/, /က်/, /tʰ/, /ခ်/, /ʧʰ/, /ဂ်/ and /ʤ/ with the similar sounds of consonant with final nasals: ⟨မ်⟩ /m/, ⟨န်⟩ /n/, ⟨ဉ်⟩ /ɲ/, ⟨င်⟩ /ŋ/, as well as the retroflex ⟨ဏ⟩ /ɳ/ and vowel sounds of /ɛ̃/, /ɔ̃/, /eĩ/, /oũ/, /iˀ/, /ɛˀ/, /aˀ/, /eiˀ/, /ouˀ/ and /auˀ/ occurred in glottal and nasal sound in different categories. Also, the phonemes of /ရွ/, /ʃ/, /ဓ/, /ð/, /ဆ/, /က်/, /tɕ/, /ခ်/, /ʧʰ/ and /ဂ်/ sounds occurred with different affricate and affricative consonant sounds included with vowel sounds of /ɛ̃/, /ɔ̃/, /iˀ/, /ɛˀ/, /aˀ/, /eiˀ/ and /ouˀ/ respectively.
