Abstract:
Determination of serum cholinesterase level by enzymatic method which has been popular for diagnosis marker for organophosphate and carbamate poisoning. The objectives of this study were to be monitoring the stability of 5,5- dithiobisnitrobenzoic acid (DTNB) solution, that is particularly important element in the reaction for serum cholinesterase level determination. Enzyme cholinesterase activity was performed using pooled serum of 6 blood donors and analyzed at day 0, day 14, day 30 and day 90 under storage temperature at 4°C of DTNB solution. This study results revealed that serum cholinesterase level at day 14 showed non-significantly compared with baseline (day 0) (p<0.05). Whereas the cholinesterase level at day 30 and day 90 showed significantly difference at p<0.05. The stability of DTNB solution in serum cholinesterase level by enzymatic method were stable for 14 days under refrigeration temperature at 4°C and this study can be applied for the preservation of the reagent solution under optimal conditions.
