Abstract:
Zoonotic pathogens such as arboviruses comprise a significant proportion of emerging infectious diseases in humans. The role of wildlife species as reservoirs for arboviruses is poorly understood, especially in endemic areas such as Southeast Asia. This study aims to determine the exposure history of different macaque species from national parks and cities in Thailand to mosquito-borne flaviviruses and alphavirus. This study focuses on dengue virus, Zika virus and chikungunya virus by conducting serological and genomic studies in free-living macaques, genus macaca, between April 2017 and March 2019. For genomic studies, 347 plasma samples from the central, eastern and western parts and also 3 National parks of Thailand were used. Nested RTPCR were performed to detect DENV1-4 and real-time RT-PCR was used for the detection of CHIKV and ZIKV. From amplified and phylogenetics analyses, nine longtailed macaques tested positive for the DENV serotype 2, but not the sylvatic DENV family. Serological studies were performed by testing serum samples collected from 25 northern pig-tailed macaques, 33 stump-tailed macaques and 4 long-tailed macaques in 3 national parks in the central, western and southern part Thailand. For the presence of antibodies against DENV, ZIKV and CHIKV by plaque reduction neutralization assay, specific neutralizing antibodies against DENV1-4 and ZIKV were mainly found in stump-tailed macaques, whereas low levels of antibody titers were detected in longtailed macaques and pig-tailed macaques. One long-tailed macaque, captured from the south of Thailand, exhibited antibody titers against CHIKV. These results suggest the enzootic of these viruses in macaques in Thailand. The ecology of arboviruses cycles should be investigated in macaques.
