Abstract:
Amphiphilic random copolymer designed to be used as drug carrier were prepared via
post-polymerization modification of poly(pentafluorophenyl methacrylate) (PPFPMA) with
poly(ethylene glycol) methyl ether amine (PEG-NH2). PPFPMA was first synthesized using
reversible addition−fragmentation chain transfer polymerization (RAFT). The
pentafluorophenyl (PFP) active ester groups of the PPFPMA reacted with the PEG-NH2 and
yielded PEG-containing amphiphilic copolymer (P(PFPMA-co-PEGMAM). The composition
of PEG in the copolymer was varied from 27 to 54 and 74%. The size of micelles selfassembled
from the copolymer in water was found to increase with the increasing PEG
composition in a range of 80-600 nm. It is our on-going effort to introduce cleavable dithiol
crosslinking to the micelles via tandem post-polymerization modification of the remaining
PFP active ester groups in the copolymer with cystamine. The ability to encapsulate a
hydrophobic dye by the micelles is also explored
