Cytotoxicity and genotoxicity of the extracts from halymenia durvillei

Abstract: Halymenia durvillei (H. durvillei), a species of Madagascar marine coasts red algae, is believed to have potential for pharmacological, nutritional and cosmetic applications. However, such potentials only make sense when devoid of any adverse effects on health consequences. Thus, the aim of this work was to investigate cytotoxicity and genotoxicity of ethanolic (HD-ET), hexane (HD-HE), ethyl acetate (HD-EA), n-butanol (HD-BU), and aqueous (HD-AQ) extracts from H. durvillei. The cytotoxicity was evaluated by MTT and LDH assays in mouse areolar fibroblast (L929), mouse macrophage (RAW264.7), mouse hepatocyte (FL83B), and human keratinocyte (HaCaT) cell lines. The genotoxicity was studied by comet assay and micronucleus test in human lymphoblast (TK6) cell line. The cytotoxicity studies indicated low toxicity to non-toxicity of HD-ET and HD-AQ as they did not significantly decrease cell viability and did not significantly increase LDH leakage from four cell lines after incubation with the highest concentration (1,000 µg/ml) of extracts for 24 and 48 h. In addition, HD-ET and HD-AQ did not show genotoxicity in vitro against TK6 cells as they were no significantly changed in tail length, percentage of DNA in tail and tail moment of comet and also no significantly induction of micronuclei after TK6 cell line was treated at the examined concentration of 10–1000 μg/ml, whereas HD-HE, HD-EA, and HD-BU have moderately high cytotoxic effect in both MTT and LDH assays and genotoxic effect in both comet assay and micronucleus test at high concentrations. The HD-HE showed the lowest IC50 value in contrast with HD-EA and HD-BU. In conclusion, H. durvillei, HD-ET and HD-AQ did not show any cytotoxic or genotoxic effects in in vitro at all concentration used while HD-HE, HD-EA, and HD-BU showed moderate toxicity at high concentrations higher than 200, 200, and 500 μg/ml, respectively. Further investigation using in vitro assays are needed to ensure safety of H. durvillei extracts in animal and humans.
