Abstract:
This work aimed to study inhibitory effect on polyphenol oxidases to quantify ascorbic acid in real sample. The PPO extracted under 3 different conditions from banana Musa acuminata Gros Michel including crude PPO, partial purification by acetone, and ammonium sulfate addition. Crude enzyme extraction was done by a homogenizer filter and a centrifuge. Consequently, a clear solution was partially purified. Catechol has been used as a substrate, Ec 1.14.18.1, for recognizing enzyme activities by optical density. The results show that the specific activities of both PAce-PPO and PAm-PPO increased about 3-6 times when compared to those of C-PPO. However, C-PPO could oxidize CAT more rapidly than the other two as determined by Lineweaver Burk plot with Km and Vmax values for CAT at 9.5 mM and 0.009 s-1, respectively. In addition, enzyme inhibitors were also studied with the additions of ascorbic acid (AA). The results showed that AA exhibited as high inhibitory effect as 97 percent on C-PPO. Inhibitory effect on C-PPO also provides linearity in range of 50-100 uM AA. The AA determination in Vit.C injection from local pharmacy was quantified by calibration curve technique that demonstrates high precision with less than 8 percent error
