Abstract:
Haloperoxidases are group of enzyme that catalyze the peroxidative halogenation reaction in the presence of hydrogen peroxide, a halide ion and a halogen acceptor. These enzymes can be classified into 3 major groups according to the range of halide ions that they utilize, chloroperoxidase, bromoperoxidase and iodoperoxidase, respectively. Iodoperoxidase utilizes iodide ion while bromoperoxidase can additionally use bromide ion. Chloroperoxidase usually accepts chloride, bromide and iodide ions as substrates. In this group, the most popular enzyme applied in coupled enzyme assays and enzyme immunoassays is horseradish peroxidase. Chloroperoxidase from Caldariomyces fumago has been extensively studied for several applications as a synthetic tool and as an analytical diagnostic reagent. Thai seaweeds naturally grown along the seacoast of the Gulf of Thailand contain bromoperoxidase activity. Bromoperoxidase from the seaweed Gracilaria changii was partially purified using 50 % ethanol precipitation and DEAE cellulose chromatography. The product was purified 20.11 fold to yield a specific activity of 12.27 U/mg protein. Chloroperoxidase from C. fumago was also partially purified using DEAE cellulose chromatography. This product was purified 4.17 fold to yield a specific activity of 1079.3 U/mg protein. In studies of analytical applications of haloperoxidases, a spectrophotometric method for measuring bromide ion concentration was developed. The assay reaction was based on the change of color from yellow to purplish blue through the bromination of phenol red to bromophenol blue catalyzed by bromoperoxidase, chloroperoxidase and lactoperoxidase in the presence of bromide ion and hydrogen peroxide. The assay method using bromoperoxidase as the brominating enzyme gave an assay range between 25 - 400 µM and with % recovery and % coefficient of variation ranges of 96.4 - 101.1 % and 1.48 - 2.65 %, respectively.

ฮาโลเปอร์ออกซิเดสเป็นกลุ่มเอ็นไซม์ที่ทำหน้าที่ เร่งปฏิกริยาการเติมหมู่ฮาไลด์เข้าไปในสารตัวรับหมู่ ฮาไลด์ที่เป็นสารอินทรีย์ โดยมีไฮโดรเจนเปอร์ออกไซด์ ร่วมด้วยเอ็นไซม์ ในกลุ่มนี้แบ่งออกได้เป็น 3 กลุ่มใหญ่ ๆ ตามความสามารถในการใช้หมู่ฮาไลด์ คือ คลอโรเปอร์ ออกซิเดส สามารถใช้คลอไรด์ โบรไมด์และไอโอไดด์ ส่วนโบรโมเปอร์ออกซิเดสสามารถใช้โบรไมด์และไอโอไดด์ สำหรับไอโอโดเปอร์ออกซิเดสสามารถใช้ไอโอไดด์ได้เพียง อย่างเดียว หลายทศวรรษที่ผ่านมา มีความพยายามนำเอาเอ็นไซม์ ในกลุ่มนี้มาประยุกต์ใช้งาน แต่ส่วนใหญ่จำกัดอยู่ที่การใช้ เอ็นไซม์ฮอสเรดิสเปอร์ออกซิเดสเอ็นไซม์อีกชนิดหนึ่งที่มี การศึกษาเพื่อประยุกต์ใช้งานคือคลอโรเปอร์ออกซิเดสที่ เตรียมได้จากเชื้อรา Caldariomyces fumago เนื่องจาก มีการสำรวจพบเอ็นไซม์ในสาหร่ายทะเลสีแดงที่ขึ้นกระจาย อยู่ตามชายฝั่งทะเลรอบอ่าวไทย ที่สามารถแสดงปฏิกริยา แบบเดียวกับโบรโมเปอร์ออกซิเดส โดยเฉพาะในสาหร่าย ทะเลสีแดงชนิด Gracilaria changii จึงอาจนำมา ประยุกต์ใช้งานในรูปแบบต่าง ๆ ได้ วิทยานิพนธ์นี้มีจุดประสงค์เพื่อเตรียมเอ็นไซม์ โบรโมเปอร์ออกซิเดส และคลอโรเปอร์ออกซิเดสให้มีความ บริสุทธิ์ขั้นต้น และนำมาศึกษาการประยุกต์ใช้ในเชิงการ วิเคราะห์เพื่อการวินิจฉัย โบรโมเปอร์ออกซิเดสจาก G. changii สามารถเตรียม ให้บริสุทธิ์ขั้นต้นด้วยการตกตะกอนโปรตีนด้วย 50 % เอทธานอล และการใช้ DEAE cellulose chromatography ซึ่งจะได้ผลผลิตที่มีความบริสุทธิ์เพิ่มขึ้น 20.11 เท่า และมี specific activity เท่ากับ 12.27 ยูนิต/มิลลิกรัม โปรตีน สำหรับการเตรียมคลอโรเปอร์ออกซิเดสให้มีความ บริสุทธิ์ขั้นต้นจาก C. fumago โดยใช้ DEAE cellulose chromatography จะได้ผลผลิตที่มีความบริสุทธิ์เพิ่มขึ้น 4.17 เท่า และมี specific activity เท่ากับ 1079.3 ยูนิต/ มิลลิกรัมโปรตีน ในการศึกษาโบรโมเปอร์ออกซิเดส คลอโรเปอร์ออกซิเดส และแลคโตเปอร์ออกซิเดส เพื่อนำมาประยุกต์ใช้ในงาน วิเคราะห์หาปริมาณโบรไมด์อิออนในสารละลายตัวอย่าง พบว่า เมื่อใช้สารฟีนอลเรดเป็นสับเสตรทที่ให้สีเมื่อเกิดการเติม หมู่โบรไมด์ แล้วโบรโมเปอร์ออกซิเดสสามารถวิเคราะห์ ปริมาณโบรไมด์ได้ในระดับความเข้มข้นที่ 25 - 400 ไมโครโมล่าร์ ด้วยความถูกต้อง และแม่นยำระหว่าง 96.4 - 101.1 % recovery และ 1.48 - 2.56 % coefficient of variation สำหรับแลคโตเปอร์ออกซิเดส ให้ผลในการวิเคราะห์ที่มีความ ไวต่ำกว่าโบรโมเปอร์ออกซิเดส ส่วนคลอโรเปอร์ออกซิเดส สามารถใช้วัดปริมาณโบรไมด์ในระดับเป็นมิลลิโมล่าร์เท่านั้น
