Culture conditions, inoculum production and domestication of wild tropical strains of Laetiporus sulphureus

Abstract: Laetiporus sulphureus (Agaricomycetes, Polyporales, Fomitopsidaceae) is widely distributed in tropical countries. L. sulphureus, an edible wood-rotting basidiomycete found on decaying wood and trunks of many tree species, produces relatively large striking yellowish or orange bracket-like fruitbodies. In this study, two L. sulphureus strains (MFLUCC 12-0546 and MFLUCC 12-0547) collected from Doi Mae Salong, Chiang Rai, Thailand, were studied in terms of nutritional composition, mineral content, optimal conditions (i.e., media, pH, and temperature) for mycelial growth, antibacterial and antioxidant activity, and possible domestication. Proximate analysis data revealed that the protein, fat, crude fibre and carbohydrate concentrations of the mushrooms were 6.91, 1.70, 6.43 and 71.74% dry weight, respectively. Their mineral contents were calculated as mg/kg dry weight for Fe (2.28), Zn (1.20), Mn (0.35), Cu (0.14), Na (11.01), Ca (2.59) and Mg (1.09). For optimal conditions of mycelial growth, potato dextrose agar and malt extract agar were favourable medium, whereas the optimum pH and temperature were 6–8 and 25–30°C, respectively. Antibacterial and antioxidant activities of the mushrooms were also investigated using disc diffusion assay and an ABTS antioxidant assay. The ethyl acetate extracts were prepared using the supernatants, derived from the mushroom mycelia, and used for both assays. Ethyl acetate extracts could inhibit the growth of all tested bacteria (i.e., Staphylococcus aureus, Escherichia coli, and Micrococcus luteus). Based on the ABTS assay system, the IC50 values of these extracts were between 0.125 – 0.170%, compared to 0.082% which was that of the Trolox standard. In addition, an attempt was made to study if the fruiting body of the Thai L. sulphureus could be formed using log and sawdust substrate. However, no fruiting bodies were produced. Only primordia developed after complete mycelial colonization of bag substrates.
