Abstract:
งานวิจัยนี้มีวัตถุประสงค์เพื่อศึกษาผลการเสริมกรดแกมมาอะมิโนบิวทีริกที่ระดับแตกต่างกันในอาหารสำเร็จรูป ต่อการเจริญเติบโต อัตราการรอดตาย และคุณภาพลูกกุ้งขาวแวนนาไม ระยะโพสลาร์วา โดยแบ่งการทดลองเป็น 4 การทดลอง การทดลองที่ 1 ศึกษาฤทธิ์การต้านอนุมูลอิสระและฤทธิ์การต้านทานเชื้อแบคทีเรียก่อโรคของกรดแกมมาอะมิโนบิวทีริกที่ได้จากเชื้อแบคทีเรียและสารสกัดพืช พบว่า กรดแกมมาอะมิโนบิวทีริกที่ได้จากสารสกัดจากพืชมีประสิทธิภาพการเป็นสารต้านอนุมูลอิสระสูง และมีความสามารถต้านทานเชื้อแบคทีเรีย Vibrio parahaemolyticus AHPND ได้ที่ความเข้มข้น 50 มิลลิกรัมต่อมิลลิลิตร การทดลองที่ 2 ศึกษาการเสริมกรดแกมมาอะมิโนบิวทีริกจากพืชซึ่งคัดเลือกจากผลการทดลองที่1 ทำการทดลองที่ระดับ 0 (กลุ่มควบคุม), 0.2 และ 0.4 เปอร์เซ็นต์ โดยคลุกกับอาหารสำเร็จรูปที่มีโปรตีน 39 เปอร์เซ็นต์ และไขมัน 7 เปอร์เซ็นต์ แล้วเคลือบด้วยพูลลูแลน ศึกษาผลต่อการเจริญเติบโต อัตราการรอดตาย และคุณภาพลูกกุ้งขาวแวนนาไม ระยะโพสลาร์วา PL13-PL25 พบว่า กุ้งขาวแวนนาไม ระยะ PL25 ที่ได้รับการเสริมกรดแกมมาอะมิโนบิวทีริก 0.4 เปอร์เซ็นต์ ในอาหาร มีความยาวตัวเฉลี่ย น้ำหนักเฉลี่ย และคุณภาพของกุ้งมีค่าสูงที่สุด ซึ่งมีค่าเฉลี่ยแตกต่างกับกลุ่มควบคุมอย่างมีนัยสำคัญทางสถิติ (P<0.05) อัตราการรอดตายเมื่อสิ้นสุดการทดลอง และอัตราการรอดตายต่อความเครียดที่เกิดขึ้นอย่างฉับพลัน มีค่าแตกต่างกันอย่างไม่มีนัยสำคัญทางสถิติ (P>0.05) และไม่พบการติดเชื้อก่อโรคตายด่วนและโรคตัวแดงดวงขาว แต่พบการติดเชื้อ EHP ในกุ้งระยะ PL9-PL25 การทดลองที่ 3 วางแผนการทดลองแบบแฟกทอเรียล 2x3 เพื่อศึกษาผลของการเสริมกรดแกมมาอะมิโนบิวทีริกที่ระดับ 0 (กลุ่มควบคุม), 0.2 และ 0.4 เปอร์เซ็นต์ในอาหาร ต่อคุณภาพของกุ้งระยะ PL25 ที่ได้รับความเครียดจากการขนส่งด้วยอัตราความหนาแน่น 100 และ 500 ตัวต่อลิตร เป็นเวลา 3 ชั่วโมง พบว่า ระดับการเสริมกรดแกมมาอะมิโนบิวทีริก 0.4 เปอร์เซ็นต์ในอาหารมีผลให้คุณภาพกุ้งและอัตราการรอดตายหลังขนส่งที่ความหนาแน่น 100 ตัวต่อลิตร สูงกว่าที่ความหนาแน่น 500 ตัวต่อลิตร โดยมีค่าเฉลี่ยแตกต่างกันอย่างมีนัยสำคัญทางสถิติ (P<0.05) การทดลองที่ 4 ศึกษาการเจริญเติบโตของกุ้งหลังขนส่ง โดยใช้กุ้งกลุ่มควบคุมที่ขนส่งด้วยความหนาแน่น 500 ตัวต่อลิตร เลี้ยงด้วยอาหารเสริมกรดแกมมาอะมิโนบิวทีริก 0, 0.2 และ 0.4 เปอร์เซ็นต์ เป็นเวลา 60 วัน พบว่า กุ้งระยะ PL27-PL87 ที่ได้รับการเสริมด้วยกรดแกมมาอะมิโนบิวทีริกระดับ 0.4 เปอร์เซ็นต์ในอาหาร มีน้ำหนักเฉลี่ยสูงสุด และเมื่อทดสอบความต้านทานเชื้อ V. parahaemolyticus AHPND แบบแช่ นาน 12 วัน พบว่า การเสริมกรดแกมมาอะมิโนบิวทีริกมีผลให้มีปริมาณเชื้อ Vibrio sp. ในตับและตับอ่อนมีค่าต่ำที่สุด และมีอัตราการรอดตายสูงที่สุดอย่างมีนัยสำคัญทางสถิติเมื่อเทียบกับกลุ่มควบคุม (P<0.05) ดังนั้น การเสริมกรดแกมมาอะมิโนบิวทีริก ระดับ 0.4 เปอร์เซ็นต์ในอาหารกุ้งขาวแวนนาไม ช่วยเพิ่มการเจริญเติบโต เพิ่มคุณภาพของกุ้งขาวแวนนาไม ระยะโพสลาร์วา ส่งเสริมความทนทานต่อความเครียดในสภาวะที่มีการเปลี่ยนแปลงความเค็มและความทนทานต่อฟอร์มาลิน ช่วยส่งเสริมคุณภาพกุ้งภายใต้ความเครียดในการขนส่ง โดยยังคงมีคุณภาพสูงกว่า 90 เปอร์เซ็นต์ ตามเกณฑ์ประเมินของกรมประมง และเพิ่มอัตราการรอดตายในสภาวะความเครียดจากเชื้อก่อโรค รวมถึงเติบโตดีขึ้นแม้อยู่ภายใต้สภาวะการติดเชื้อ EHP. The study aimed to investigate the effects of different levels of gamma-aminobutyric acid (GABA) supplementation in
Pacific white shrimp (Litopenaeus vannamei) diet on growth performance, survival rate, and postlarvae shrimp (PL) quality. The study
was divided into 4 experiments. The first experiment was focused on the antioxidant properties and antibacterial activity of GABA from
lactic acid bacteria and herb extracts. The results found that GABA from herb extracts had high antioxidant properties and were resistant
to Vibrio parahaemolyticus AHPND bacteria at a concentration of 50 mg/ml. The second experiment was investigated on the effects of 0%
(control), 0.2%, and 0.4% GABA, result from the first experiment by supplementation on top to a commercial diet of 39% CP and 7% CF
then coated with pullulan, on growth performance, survival rate, and postlarval quality PL13-PL25. The results found that PL25 shrimp
fed 0.4% GABA had the highest average length, average weight, and postlarval quality with statistically significant differences (p<0.05)
from the control group (0%). The survival rate at the end of the experiment under normal condition and under salinity and formalin stress
conditions were not significantly different (p>0.05). In all experimental groups, there were not found the infection by a pathogen caused
early mortality or white spot disease. On the contrary, an EHP infection was found in the hepatopancreas of PL9-PL25 shrimp. The third
experiment was assigned in factorial of 2X3 in CRD. The experiment was investigated the effects of 0% (control), 0.2%, and 0.4% GABA
on PL25 shrimp quality after high stocking density transportation of 100 and 500 individuals per liter for 3 hours. The results found that
the postlarval shrimp quality and survival rate after transport at a density of 100 individuals per liter was higher than 500 individuals per
liter and shrimp quality certainly related to levels of 0.4% GABA in the diets with statistically significant differences (p<0.05). The fourth
experiment was study on growth performance of shrimp after transportation by focusing on the control group which transportation at
density 500 individuals per liter and fed 0, 0.2, and 0.4% GABA on top to the diet for 60 days. The result found that PL27-PL87 shrimp
fed 0.4% GABA had the highest average weight (p<0.05). The disease resistance against V. parahaemolyticus AHPND by bath treatment
for 12 days demonstrated that shrimp fed 0.4% GABA had the lowest Vibriosp. count from the hepatopancreas, and the highest survival
rate with statistically significant differences (p<0.05) from the control group.
Therefore, White leg shrimp diets supplemented with 0.4% gamma-aminobutyric acid could improve postlarvae shrimp
growth performance, shrimp quality including resistance to salinity and formalin stress conditions, enhancing shrimp quality under
transportation stress with the shrimp quality over 90% according to the evaluation criteria of the Department of Fisheries,
Thailand, increase the survival rate under pathogenic stress conditions and improved growth even under EHP infection conditions
