Abstract:
การเสริมยีสต์ออโตไลเซทในอาหารต่อการเติบโตและระบบภูมิคุ้มกันของกุ้งขาว แบ่งเป็น 5 การทดลอง การทดลองที่ 1 ศึกษาการเสริมยีสต์ออโตไลเซทในอาหารต่อการเติบโตและระบบภูมิคุ้มกันในกุ้งขาวระยะโพสลาวา 28 วางแผนการทดลองแบบสุ่มตลอด (CRD) 3 ชุดการทดลองคือ อาหารเสริมยีสต์ออโตไลเซท 0, 0.1, และ 0.2% ผลการศึกษาพบว่าการเสริมยีสต์ 0-0.2% ไม่มีผลต่อการเติบโต (p>0.05) แต่กุ้งที่ได้รับยีสต์มีกิจกรรมฟีนอลออกซิเดสและซูเปอร์ออกไซด์ดิสมูเตสที่สูงกว่าชุดควบคุมอย่างมีนัยสำคัญ (p<0.05) เมื่อเลี้ยงในสภาวะที่มีแอมโมเนีย กุ้งขาวที่ได้รับยีสต์ออโตไลเซทมีซูเปอร์ออกไซด์ดิสมูเตสสูงกว่าชุดควบคุม (p<0.05) เมื่อเคลือบเชื้อวิบริโอพาราฮีโมไลติคัสบนอาหาร กุ้งขาวที่ได้รับยีสต์ออโตไลเซทมีโปรตีนในเลือดและกิจกรรมฟีนอลออกซิเดสที่สูงกว่าชุดควบคุม (p<0.05) การทดลองที่ 2 ศึกษาการเสริมยีสต์ออโตไลเซทในอาหารต่อการเติบโตและระบบภูมิคุ้มกันในกุ้งขาวระยะเติบโต วางแผนการทดลองแบบสุ่มตลอด (CRD) 3 ชุดการทดลองคือ อาหารเสริมยีสต์ออโตไลเซท 0, 0.1, และ 0.2% พบว่า การเสริมยีสต์ที่ 0-0.2% ไม่มีผลต่อการเติบโต (p>0.05) แต่กุ้งที่ได้รับยีสต์มีปริมาณเม็ดเลือดสูงกว่าชุดควบคุมอย่างมีนัยสำคัญ (p<0.05) เมื่อฉีดเชื้อวิบริโอพาราฮีโมไลติคัส กุ้งขาวที่ได้รับยีสต์ออโตไลเซทมีซูเปอร์ออกไซด์ดิสมูเตสที่สูงอย่างมีนัยสำคัญ (p<0.05) ประสิทธิภาพการย่อยได้ของอาหารที่เสริมยีสต์ออโตไลเซทร่วมกับเอนไซม์ของกุ้งขาว (in vitro) พบว่าการเสริมยีสต์ออโตไลเซทที่ระดับ 0.1-0.2% มีผลต่อค่าการย่อยได้ของกรดอะมิโน โปรตีน มอลโทส ไซโลส กลูโคส แมนโนส และน้ำตาลรวม กุ้งขาวทีได้รับยีสต์ออโตไลเซทมีเม็ดไขมันและความสมบูรณ์ของตับมากกว่าชุดควบคุม และควบคุมเชื้ออีเอชพีได้ เมื่อวิเคราะห์ด้วยวิธี Real time PCR (qPCR) การทดลองที่ 3 ศึกษาประสิทธิภาพการย่อยได้ของอาหารที่เสริมยีสต์ชนิดต่างๆร่วมกับเอนไซม์ของกุ้งขาว (enzyme assay) พบว่าการเสริมยีสต์หมัก ยีสต์แห้ง และยีสต์นิวคลีโอไทด์ในอาหารร่วมกับเอนไซม์กุ้งขาว ทำให้มีค่าย่อยได้ของโปรตีนและคาร์โบไฮเดรตดีขึ้น การทดลองที่ 4 ศึกษาเอนไซม์ในผลิตภัณฑ์ยีสต์ชนิดต่างๆ ยีสต์ออโตไลเซท ยีสต์ทั้งเซลล์แห้ง ยีสต์แห้ง และยีสต์หมัก มีเอนไซม์เซลลูเลส โปรติเอส ไลเปส อะไมเลส และโปรตีนที่แตกต่างกันอย่างมีนัยสำคัญ (p<0.05) การทดลองที่ 5 ศึกษาประสิทธิภาพการเป็นสารต้านอนุมูลอิสระในยีสต์ออโตไลเซท, ยีสต์ทั้งเซลล์แห้ง, ยีสต์แห้ง, ยีสต์หมัก และวิตามินซี โดยวิธี DPPH radical scavenging, Ferrous ion chelating, ABTS assay และ Folin-Ciocalteu method ทดลองแบบสุ่มตลอด (CRD) พบว่าผลิตภัณฑ์ยีสต์แต่ละชนิด มีความสามารถในการเป็นสารต้านอนุมูลอิสระด้วยกันทั้งสิ้น The study on the effects of supplemental yeast product in shrimp feed on growth performance and immunity of Pacific white shrimp (Litopenaeus vannamei) was conducted. The research was divided into 5 experiments. The first experiment was a study on the effects of supplemental autolysate yeast on growth performance and immunity of post-larvae 28 by assigned in a complete randomized design with 3 treatments of 0, 0.1 and 0.2% autolysate yeast on top. There was not significant difference in growth performance (p>0.05). Shrimp fed autolysate yeast 0.1-0.2% had significantly higher phenoloxidase activity and superoxide dismutase (p<0.05). After an ammonia stress test, shrimp fed autolysate yeast 0.1-0.2% had significantly higher level of superoxide dismutase (p<0.05). In the oral administration challenge test by feed coated with Vibrio parahaemolyticus EMS, shrimp fed autolysate yeast showed significantly higher hemolymph protein and phenoloxidase activity (p<0.05). The second experiment was a study on the effects of supplementation autolysate yeast on the growth performance and immunity of grow-out stage by assigned in a complete randomized design with 3 treatments of 0, 0.1 and 0.2% autolysate yeast on top. There was no significant difference in growth performance (p>0.05) but noticed a significantly higher total hemocyte count (p<0.05). After challenged by injecting V. parahaemolyticus, shrimp fed autolysate yeast 0.1-0.2% had significantly higher levels of superoxide dismutase (p<0.05). The in vitro digestibility of diets supplemental
autolysate yeast by using shrimp digestive enzyme was determined. The results showed high nutrient digestibility (p<0.05) on amino acid, protein, maltose, xylose, glucose, mannose and total sugar. The hepatopancreases of shrimp fed autolysate yeast showed plenty of oil droplets, healthy hepatopancreases and ability to control EHP. The third experiment was a study on the in vitro digestibility of the shrimp diet in combination with different yeasts by using shrimp enzyme was conducted. The results showed the high ability (p<0.05) of fermented yeast, dried yeast and yeast nucleotide on improve protein and carbohydrate digestibility (p<0.05). The fourth experiment was a study on the enzyme assay on yeast products. There were significantly higher levels of cellulase, protease, lipase, amylase, and protein in yeast products (p<0.05). The fifth experiment was a study on the effectiveness of antioxidants in yeast products by DPPH, FIC, FolinCiocalteu, and ABTS methods. The results showed that all products had antioxidant properties
