Abstract:
A broad range gas chromatography-flame ionization detector (GC-FID)-based metabolite profiling method was applied to investigate the metabolic compounds of pre- and post- Fusarium verticillioides inoculated kernels of five coloured Thai corn hybrids grown at the same location and season. Additionally, spectrophotometric methods were conducted to determine amylose and total anthocyanin content in the samples. Applied metabolite profiling, in combination with multivariate analysis, revealed increases in the majority of lipid metabolites (fraction I and II) in corn variety 7566 as well as acid metabolites (fraction IV). This variety also contained high amount of nutritionally important phytosterols (i.e. sitostanol, campesterol, stigmasterol and ∆7-campestenol). Highest anthocyanin contents were recorded in variety CP09 while amylose was significantly highest in 7566. PCA separated Fusarium verticillioides inoculated corn kernels from their pre-inoculated counterparts. Semi-quantitative analysis revealed the decrease in the majority of lipids in post-inoculated kernels. Sugar and sugar alcohols (fraction III) also increased in post-inoculated samples, except di-and trisaccharides. Majority of acid (fraction III) metabolites showed significant increase at 7 days post-inoculation time point.
